I'm using the alpha theme - great theme!

Some of my users are selecting the region / state when posting new listings instead of selecting the city. At: https://alpha.mb-themes.com/item/new in the location box, you can select country, state or city for the location. Can you make it so that the user can only select the final level - city  for the Location field?

No as i.e. US can have 50k of cities. You can start typing and all relevant values will be shown to you.

I think you have misunderstood what I was trying to say.

If a user is placing an advert and starts typing or selects one of the drop down regions, some are not selecting a city, they select a region instead. Is it possible so that the drop down selection is disabled when posting a listing, and that only cities are shown when you begin typing for the location field?

You can do such modifications but for search as well, check ajax.php in theme folder
